Description: The monoclonal antibody UP-H2 recognizes human IREM-2 (immune receptor expressed in cells derived from monocytes) also known as LMIR6, and unofficially CD300E and CD300LE. It is a member of the CMRF-35 family. This 34 kDa Ig-containing receptor is an activating receptor and, through its charged residues in the cytoplasmic domain, interacts with ITAM-containing adapter proteins such as DAP12.

Expression is found exclusively in monocytes and myeloid DCs. The expression may be transient as monocyte differentiation leads to downregulation of IREM-2, but the expression is found in mature developing countries. Myeloid cell lines such as THP-1 and U937 have also been shown not to express IREM-2. The in vitro addition of UP-H2 antibody, either soluble or cross-linked, stimulates monocytes.

Informed Applications: This UP-H2 antibody has been reported to be used in flow cytometric analysis.

Proven Applications: This UP-H2 antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. This can be used at the rate of 5 µL (0.25 µg) per test. A test is defined as the amount (µg) of antibody that stains a cell sample in a final volume of 100 µL. The number of cells must be determined empirically, but can range from 10 ^ 5 to 10 ^ 8 cells/test.

Destination information

CD300e is a member of the glycoprotein family of cell surface proteins expressed in myeloid cells. CD300e interacts with the tyrosine kinase-binding protein TYRO protein and is believed to participate as an activating receptor. CD300e is expressed on mature myeloid dendritic cells and monocytes and is down-regulated on immature dendritic cells. Additionally, CD300e interacts with DAP-12, which mediates activation signals. The alloreactive response of naïve T cells has been reported to increase with activation of CD300e. Diseases associated with CD300e dysfunction include tungiasis and lice infestation.
